Role of adrenomedullin in pulmonary tumor

OBJECTIVE:To review the document about the relationship between adrenomedullin and pulmonary tumor.METHODS:PubMed central and CHKD periodical full-text databases were retrieved with the key words of peptide,adrenomedullin,pulmonary and tumor.The 40 pertinent literature from 1993 to 2009 was screened.The inclusion criteria:1)the expression of adrenomedullin in pulmonary tumor.2)the role of adrenomedullin in regulation of tumor cell.3)the effect of adrenomedullin in tumor angiogenesis.4)the advanced research of mech-anism of adrenomedullin.Eventually,21 documents were internalized.RESULTS:Adrenomedullin(ADM)is highly expressed in various tumor tissues and cell types,including lung cancer.It is suggested that ADM may be involved in lung carcinogenesis.NSCLC cells are important targets of AM and AM may regulate activities in these malignant lung cells through differential induction of various early response genes including c-fos,c-jun and egr-1.Hypoxia is one of the characteristic microenvironments of tumor growth.The expression of ADM in lung cell is regulated by HIF-1.Targeting adrenomedullin receptors(AM-Rs)inhibits tumor angiogenesis and suppresses the growth of human tumor in mice.AM-Rs are also synthesized in vivo and in vitro by a variety of tumor cells that also express the ligand.The present findings suggest that the ADM/AM-R regulatory system plays a major role in respiratory physiology and lung carcinogenesis.CONCLUSION:The inhibition of the interaction between peptide hormones and their receptors,utilization of negative growth factors,interruption of the signal transduction pathways,or effecting decreased peptide hormones and(or)receptor expression seem logical possibilities for new and specific treatment approaches,which hopefully will translate into new,less toxic,more effective,and desperately needed lung cancer treatment.
