Experimental study on drug sensitivity of the Her-2-positive gastric carcinoma cell lines to trastuzumab

Objective To exploit the drug sensitivity of Her-2-positive gastric carcinoma cell lines to trastuzumab by in vitro experiments.Methods The expression levels of Her-2 protein of several human gastric carcinoma cell lines,including NCI-N87,SGC7901,MKN48 and MKN25,were detected by Western-blot.The cell inhibition ratios of trastuzumab were measured using MTT method.Results The Her-2 protein expression of four gastric carcinoma cell lines were positive,and the highest level was detected in NCI-N87 cells (P =0.000).When the concentration of trastuzumab were 10 μg/ml,20 μg/ml and 40 μg/ml,the inhibition ratios of NCI-N87 cell line were the highest (P= 0.000 for all) and the half inhibitory concentration (IC50) was the lowest.Cell inhibitions of the four gastric carcinoma cells increased following with the raised concentration of trastuzumab.Conclusion Trastuzumab could inhibit the growth of Her-2 positive gastric carcinoma cell lines,and the inhibition correlated with the expression levels of Her-2 protein and depended on the dosage of trastuzumab.
