Development of EST-SSRs Using Public ESTs in Hevea

More and more ESTs(expressed sequence tags) of Hevea cDNA sequence have been produced,which are a good public sequence source for development of EST-SSRs.11809 ESTs were assembled into 3932 unigenes in which 323 ESTs includes 358 SSRs,about 9.1% unigenes inclusive of SSR.There were 93 SSRs identified,of which dinucleotide repeats(57.5% of SSR-ESTs),trinucleotide repeats(26.0%) and pentanucleotide repeats(12.8%) contributed to 96.3% of the total repeats.158 primer pairs that satisfied melting temperature conditions and PCR product sizes were designed based on the flanking sequences of the 323 ESTs.60 SSRs(38%) were successfully amplified,of which 40(66.7%) showed plolymorphism in 48 accessions of germplasm including 31 accessions of Wickhanm germplasms,12 accessions of 1981' wild germplasms,2 accessions of H.nitida,2 accessions of H.spruceana and 1 accession of H.pauciflora.Development of Hevea EST-SSRs marker is beneficial to marker-assisted breeding and selection,and functional diversity analysis in Hevea.
