Analysis of Cytochrome C Oxidase I Genes of Five Species of Margarya and Cipangopaludina chinensis

HUANG Xiao-yan,WANG Li-zhen(College of Life Science,Yunnan University,Kunming650091,China)
Take several species of Margarya(Gastropoda: Viviparidae) and Cipangopaludina chinensis(control) as the object of study, the genomic DNAs in liver were extracted.Mitochondria cytochrome C oxidase I(COI) gene fragments of the snails were amplified by PCR method and then purified and sequenced, and used DNAStar package on the sequence editor, and the program MEGA was used for reconstructed phylogenetic tree with NJ and UPGMA methods respectively. The result indicated: The COI gene size is 636bp,the genealogical tree demonstrated that M.melanioides,M.melanioides dianchiensisi,M.monodi and M.tropidophora compose one groups,M.yangtsunghaiensis are far phylogenetic relationships with the above snails's,become the other group which in the genus Margarya evolves alone; The Cipangopaludina chinensis and the genus Margarya genetic distance reaches between 0.404~0.518%,the phylogenetic relationships is very far.
