The Study on Gliadin Variation of Sichuan Major Wheat Cultivars

In order to investigate the special variation of gliadin alleles and analyse the genetic difference between different genotypes, 40 cultivars which had once popularized above 1000000mu per year in Sichuan during the recent 50 years were used in this study with acid polyacrylamide gel electrophoresis (APAGE). The results were as follows: There were 38 gliadin genotypes in the 40 cultivars, with 9 cultivars bearing the allele Gli Bll, a marker of 1BL/1RS translocation. 46 gliadin bands were seperated by electrophoresis and 40 of them showed polymorphism (amount to 84 8%). The genetic distance (GD) based on gliadin ranged from 0 to 0 67, with an average of 0 33. There were relatively higher GD between the early and recent 20 years cultivars of 1BL/1RS and non 1BL/1RS translocation lines. Cluster analysis shows that the studied cultivars could be classified into 4 clusters at the level of GD 0 35, and the majority of variaties with same consanguinity are clustered together. Based on these results, it is concluded that Sichuan wheat cultivars have abundant variation and high genetic diversity.
