The application of Flow cytometry on the DNA content and ploidy analysis of hydrophily organisms

GENG Bo1, SUN Xiao-wen1 (Hei Longjiang Fishery Research Institute, Chinese Academic of Fishery Sciences,Harbin 150070,China)
We used flowcheck to determine the DNA content and analyze the ploidy of many freshwater and seawater fishes such as, Carassius auratus gibelio,Pelteobagrus fulvidraco, Paralichthys olivaceus and so on.With several hundred samples analysis, it can be think that the results of determination with FCM are stable and consistent and FCM can be used to work on a great deal of samples. On the experiments, we also to do a series of research on the sample’s conservation, deal with and the quantity of add samples to find the best way of conservation and the easiest deal measure with samples and the lowest quantity. FCM has important roles on the genetic breeding, stock differentiation of hydrophily organisms.
