STUDY ON INDUCTION OF EMBRYOGENIC CALLUSE AND PLANTLET REGENERATION IN MAIZE

Embryogenic maize calli were inducted from the immature embryos of commercially important genotypes.From these calli plantlets were regen- erated.Under suitable conditions,the induction frequency of embryogenic calli from Yuanqi722×5003 and other genotypes was 90%,the frequency of calli differentiating buds was more than 80%.The buds grew up robust plantlets on the modified N_6 medium supplemented with 1 mg/L 6-BA and 0.1% activated carbon.The size and genotypes of the immature embryos inoculated,basic media and sucrose concentration,the components and concentration of phytohormones significantly affected the induction,subc- ulture and differentiation of the embryonic calli.The induction frequency of embryogenic calli could be improved obviously if immature embryos cultured for 7 or 8 days were clamped into pieces.In regenerated plantlets the frequency of aberrant ones increased as the embryogenic calli were repeatedly cultured.
