Determination of Cholesterol in Edible Oil by Gel Permeation Chromatography-High Performance Liquid Chromatography

Objective To establish the method for determination of cholesterol in edible oil by high performance liquid chromatography(HPLC) coupled with gel permeation chromatography(GPC). Methods Edible oil was dissolved in ethyl acetate-cyclohexane and then purified by GPC with a flow rate 5.0 m L/min. The portions between 18-24 min were collected then separated on a C18 column and detected by a diode array detector in 205 nm. Results The linear range of cholesterol was 0.05-0.8 mg/m L, the limit of detection was 7.07 mg/kg. The recoveries were from 69.96 % to 93.87 %, and the relative standard deviations(RSDs) were from 0.38 % to 6.76 %. Conclusion The pretreatment of this method is easy-operating and high automatic, thus it is suitable for detection of cholesterol in edible oil.
