Study on dose of steep pulsed electric fields in rabbit liver

Objective To explore dose-effect relationship between the electric field therapeutic dosage and the casualty effect in rabbit liver in vivo.Methods Forty-eights rabbits were divided into 3 groups i.e.voltage group(n=21),pulse width group(n=12) and frequency group(n=15).The rabbit livers in vivo were exposed to steep pulsed electrical fields with serial dosage,then the rabbits were sacrificed after 72 hours,and necrosis area of livers were surveyed.Results In voltage group 21 rabbit livers in vivo were exposed to steep pulsed electrical fields with voltage of serial dosage 400 V,500 V,600 V,700 V,800 V,900 V,1 000 V,which was fixed pulse width of 10 μs,frequency of 20 Hz in 10 minutes,necrosis area of livers were(2.00 ± 0.00) mm2,(6.67 ± 2.08) mm2,(9.67 ± 2.08) mm2,(41.67 ± 2.89) mm2,(149.33 ± 5.03) mm2,(153.00 ± 4.36) mm2,(157.33 ± 2.08) mm2 respectively.In pulse width group,12 rabbit livers in vivo were exposed to steep pulsed electrical fields with pulse width of serial dosage 0.5,1.0,5.0,10.0 μs ,which was fixed voltage of 800 V,frequency of 20 Hz in10 minutes,and necrosis area of livers were(0.00 ± 0.00) mm2,(6.33 ± 1.53) mm2,(40.67 ± 2.52) mm2,(149.33 ± 5.03) mm2 respectively.In frequency group,15 rabbit livers in vivo were exposed to steep pulsed electrical fields with frequency of serial dosage 20 Hz,40 Hz,60 Hz,80 Hz,100 Hz ,which was fixed voltage of 800 V,pulse width of 10 μs in 10 minutes,necrosis area of livers were(149.33 ± 5.03) mm2,(152.00 ± 2.00) mm2,(154.00 ± 2.00) mm2,(162.67 ± 3.21) mm2,(173.33 ± 2.89) mm2 respectively.The rabbit livers in vivo were exposed to electrical pulsed fields of serial dosage and liver necrosis area increased gradually with increasing of electric-field intensity.Conclusion It is demonstrated a clear dose-effect relationship between necrotic area and steep pulsed electric fields,its value increases along with the dosage increasing.
