Combined Effects of Temperature and pH on Hatchability of Fertilized Egg of Sepia pharaonis by Using Response Surface Method

On the basis of one-factor tests and the central composite design(CCD) principle, the method of response surface analysis was adopted to evaluate the combined effects of temperature(24–32 ℃) and pH(7.0–8.5) on hatchability of Sepia pharaonis. The results showed that the linear and quadratic effects of temperature and pH had a very significant effect on hatchability(P 0.01).The interactive effect between temperature and pH also had a very significant effect(P 0.01). Hatchability increased and then decreased as the temperature or pH increased. The model equation of hatchability(Y) towards temperature(X_1) and pH(X_2) were established as follows: ? = 85.27–6.74 X_1 + 5.22 X_2 + 9.42 X_1~2–12.43 X_1~2–8.18 X_2~2(R2 = 0.978 6; adjusted R2 = 0.963 3; Adeq precision = 22.974). The experimental results indicated that the proposed model was reasonably accurate and can be used for describing and predicting the hatchability. Solving the regression equation for temperature equals to 27.5 ℃ and pH equals to 7.9, the optimum predicted value for hatchability was 86.46% and the reliability peaked to 0.949. Moreover, since verification result of optimum conditions accorded with the predicted value, it is suggested that the hatching of fertilized egg of S. pharaonis can be improved by rearing at an optimal combination of temperature and pH.
