IDENTIFICATION OF HEAT TOLERANCE AND SELECTION OF HEAT TOLERANT VARIANTS FROM B. campestris VAR. Chinensis iN VITRO CULTURE

Seed germination, seedling growth and hypocotyl protoplast vigor of B. campestris var.chinemsis were inhibited by 35-40℃temperatures in vitro culture. The heat tolerance of germinating seeds for the tested cultivars with low heat tolerance was obviously improved by 25℃ pre-culture treatment and seed soaking with 1 500 mg·L(-1) Hyp.The heat-tolerant levels of seedlings were able to be increased by 3% sucrose as well as 34℃or 0℃ pre-culture treatment,the increase for the tested cultivars with low heat tolerance was more obvious than that for the tested cultivars with higher heat tolerance.Variants with higher heat tolerance were obtained by the methods of soaking seeds with pingyangmycin and then germinating seeds and culturing seedlings at higher temperatures.This variance was able to be transferred to progenies through vegetative propagation way.
