EXPERIMENTAL IMMUNOTHERAPY OF HUMAN GASTRIC CANCER IN NUDE MICE WITH LYMPHOKINE-ACTIVATED KILLER CELLS INDUCED BY rIL-2

In this study rIL-2 was used to induce mouse spleen cells or human PBMO to transform intoLAKcells at the concentration of 1,000 u/ml in a co-culture system for 6 days, which had been shown in our previous work as the optimal condition to get satisfied anti-tumor effect in vitro, A modified schedule (the 3×5 plan) was developed to assess the effect of the Chinese made rIL-2 on cancer immunotherapy. When the tumors transplanted into mice entered their rapid growing stage on the fifth day after subcutaneous inoculation,rIL-2 was administrated intraperitoneally for 5 days at the dosage of 1,000 u aday for each mouse, followed by another five days during which the rIL-2 was given at the same dosage together with 10 LAK cells. Then the third 'five days' began in which rIL-2 was injected alone again, allowing treatment of the transplanted tumor in vivo for consecutive 15 days. judged by changes of tumor size and weight, the inhibitory indices were 68.7%for mouse liver tumor HAC in Balb/c mice and 52.4%for solid tumor of human,gastric cancer cell line 7901 in nude mouse respectively. It was evident that our adoptive immunotherapy in the above regimen was safe to the tumor-bearing animals and quite effective in suppression of tumor growth.
