A study on the solid culture and submerged liquid Culture for Bacillus mucilaginosus

WU Hong-sheng,GUO Xun-bing (Yangzhou AgriculturalResearch institute - Yangzhou Jiangsu Province, 225002,China)
General culture characters and formulations of submerged liquid culture of the Bacillus mucilaginosus strain K906 isolated from soil in Yangzhou are discussed in the paper. It showed that strain K906 produces capsule in the nitrogen-free solid sulture medium while having no capsule but much more rod-shape bacteria and spores in the nitrogen medium. Also, the strain grows much faster in the N-medium than in the N-free medium, about 10 times, the strain uses the xoluble starch as carbon source and be grown much better in the addition of yeast powder in the medium that maximum counter of the bacteria in flask-shaking culture gets up to 10. 0x108 bacterium/ml in 3-4days. Eight batches trail industrial fermenta- tion of the soluble starch amonia submerged liquid culture in the tank of 1000L. harvest an average counter of 18. 9 × 108bacterium/ml with formulation period of 40- 60hrs suitable for large-scale production.
