Isolation and characterization of an acidic polysaccharide from Agrimonia pilosa Ledeb

Purpose To study the isolation and purification of polysaccharides from Chinese herb Agrimonia pilosa Ledeb.Methods Cellulose DE-52 and Superdex-200 column chromatography were used to isolate and purify the extract of polysaccharides.Physicochemical Properties of the polysaccharides were studied by GC,FTIR,GPC analysis.Results A fraction HAPⅢ was obtained.Detection showed that the content of total polysaccharides,protein and uronic acid in HAPⅢ was 81%,3.4% and 45% respectively.GC analysis indicated that HAPⅢ was composed of Gal,Glc,Rha,Ara,Man,Fuc and Xyl with a molar ratio of 5.65∶2.67∶2.61∶1.86∶1.12∶1.09∶1.The average Mr was estimated to be 1.90×105 by gel filtration chromatography.Conclusion HAPⅢ is an acidic polysaccharide.
