Expression of ERK and TrkB in rat cortical astrocytes

We investigated the expression of ERK(extracellular signal-regulated kinase)and trkB(tyrosine kinase receptor B)in cultured rat cortical astrocytes.The brains were removed from postnatal 2-3 days SD rats under sterile condition.Cell suspension was prepared by digesting the tissue with trypsin.The purity of astrocytes was identified using glial fibrillary acidic protein(GFAP)immunocytochemistry.The expression of TrkB,ERK1 and ERK2 protein were determined by immunocytochemistry and Western blot analysis.The results showed that the purity of astrocyte was more than 95% by GFAP immunocytochemistry.ERK1 immunostaining was observed in cytoplasm of the astrocytes,and TrkB immunopositive staining was located in cytomembrane,cytoplasm and nucleus.The bands of ERK1 and ERK2 were clearly found after development with enhanced chemiluminescence(ECL).These results indicate that rat cortical astrocytes express TrkB,ERK1 and ERK2,and TrkB/ERK signal transduction pathway may be related to proliferation of astrocytes.
