EXPERIMENTAL STUDY ON GAP-43 EXPRESSION IN CONDITIONING LESION AND REGENERATION OF RAT SCIATIC NERVE

It bas been reported that GAP-43 (growth-associated protein) is a neuron-specific protein which has been found and studied for 10 years or more. Although it might play an important role in neuronal development and regeneration, its real function is unclear; therefore it is necessary to study further. The purpose of this study is to explore Gab-43 expressions during nerve injury and regeneration using immunohistochemical technigues. GAP-43 expressions have been observed and analysed after sciatic nerve transection and epineurium suture, squeeze (or crush) and re-squeeze lesions in 60 rats. The main results are as follows: 1. GAP-43 peitive immunoreactivity appeared in both the cell body (the lumbar spinal cord and dorsal root ganglia) after sciatic nerve trsusectiou or squeeze injury, but in the proximal stumps the fibers became strohg poritire on. week after injury and in the distal stumps two weeks after ihjury.2. GAP-43 positive immunoreactivity was weakened in all parts of the petipheral neurons oaf month postoperatively ;and was nearly normal two months postoperatively .3. The positive reaction appeared again in re-squeeze injury group thirty and sixty day. postoperatively .4. Positive immuooreactiou was weakehed gradually in the spinal cord of the injury side, and the localization of reaction is in laminae Ⅶ,Ⅷ and Ⅸ of the ventral horn.The results of GAP-43 immunohistochamical staining demonstrated that the immunostainiug in the CNS was weakened, and in PNS it was strengthened from the proximal to the distal stumps gradually and then returned to normal. The results show that GAP-43 is syhthesised mainly in the body of neuron and transported along axon to the site of injury and regeneration. It has been suggested that the Guy-43 takes part in neuronal regeneration and play. a critical role in axonal growth and regeneration directly or indirectly. Gab-43 immunohistochemical stain can also be used for evaluating tea neuronal regeneration.
