Effects of 2-PAM on acute lung injury induced by PFIB inhalation in mice

Objective To study the effects of 2-PAM on acute lung injury induced by perfluoroisobutylene (PFIB).Method PFIB concentration and its chemical transformation efficiency with 2-PAM were determined by gas chromatography,and the total protein concentration in bronchoalveolar lavage fluid(BALF)and lung wet/dry weight ratio were also determined at 24th hour after inhalation of PFIB,meanwhile,the effects of 2-PAM on mice before or after PFIB exposure were observed,too.Result The preventive administration of 2-PAM could greatly lower the wet/dry ratio of lung-to-body weight in mice,the survival ratio was also risen in this group,and its chemical transformation efficiency was 64.8%.It was suggested that PFIB should be easy to combine with hydroxyimino ion,2-PAM might reduce the toxicity of PFIB.Therefore,it could be considered that the good effect of 2-PAM might be related to its neuclophilic property.Conclusion 2-PAM could significantly suppress the leakage of protein in lung,reduce the wet/dry ratio of lung-to-body weight yatio and elevate the survival rate of PFIB inhaled in mice,thereby has a definite preventive effect on ALI caused by PFIB inhalation.
