Production, Optimization and Properties of Exo-nulinase by Aspergillus ficuum SK004

Peng Yingyun Jiang Bo Jin Zhengyu(The Key Lab of Food Science and Safety, Ministry of Education, Southern Yangtze University, Wuxi, 214036,China)  
A strain of Aspergillus ficuum SK004 which produces extracellular exo-inulinase was stored in our laboratory. The optimal medium compositions. were (g /L): inulin 25, peptone 25, NaCl 5, NH 4H 2PO 4 4, MgSO 4 0.5, ZnSO 4 0.1, pH 6.5.The cultural conditions were temperature 30℃, shaken at 200 r/min with 30*#mL medium per flask (250*#mL) for 7.5 d. The maximum enzyme activity was 53.1 U /mL. The optimal reaction temperature and pH of exo-inulinase were 60℃ and 4.5 respectively. The enzyme was stable within pH 3.0~8.0 under 60℃.
