Study on ultrasonic-assisted extraction and activity of scavenging DPPH free radicals of total flavonoid from Toona sinensis (A.Juss.) roem leaves

A mothed ,namely ultrasonic-assisted extraction ,was used to extract total flavonoid from Toona sinensis(A.Juss.)Roem leaves .The optimum extraction process was optimized with orthogonal test .Experiments were made by using L_16(45) orthogonal design with extractive yield of total flavonoids as the first index,IC_50 of scavenging DPPH free radicals of total flavonoids as the second index, investigating the influence of ethanol concentration, extracting temperature, extracting time , the ratio of solid to liquid and extracting times. Optimum extraction conditions were determined as follows: temperature 50 ℃,time 60 min, 70% EtOH as extracting solvent ,stock ratio 1∶10 (g/mL), ultrasonic-assisted extracting for four times. The extractive yield of total flavonoids from Toona sinensis(A.Juss.)Roem leaves was 22.2134 mg/g. IC_50 of scavenging DPPH free radicals of total flavonoid was 19.1327as the optimum parameters.
