Study on extraction,purification and component of polysaccharides from Allium macrosttemon Bge

XIA Xin-kui,YANG Hai-xia,LI Chun,ZHOU Wei(Department of Biotechnology,Xinyang Agricultural College,Xinyang 464000,China)  
The polysaccharides from Allium macrosttemon Bge.were separated,purified by DEAE-cellulose chromatography and Sephadex G-150 gel chromatography.Their purity were tested by cellulose acetate pellicle electrophoresis and gel filtration on Sephadex G-150.Their component were measured by means of thin-layer chromatography(TLC).The result showed:PAM was separated into three fractions(PAM-I,PAM-II and PAM-III)by DEAE-cellulose chromatography.Then they were purified by Sephadex G-150 gel chromatography into PAM-Ib,PAM-IIa and PAM-III'.The identification of purity implied that they were homogeneous components.TLC result showed PAM-Ib was composed of Galactose and Glucose.PAM-IIa was consisted of Galactose,Glucose,Fructose,Xylose and Rhamnose.The main compositions of PAM-III' were Galactose,Glucose and Xylose.
