Optimization of fermentation medium for thermophilic fungal xylanase and study on some enzymatic properties

Fermentation medium of thermophilic fungal for xylanase production was optimized by response surface method and the basic properties of the enzyme solution were studied. First,eight kinds of factors of the initial medium was adopted by the Plackett-Burman design,and selected three kinds of important factors,namely,corn cob,urea and KH2PO4,and then the path of steepest ascent was adopted for the central composite experiment to determine the maximum response interval. Finally response surface optimization results was obtained,corncob 4.570%(w/v) ,urea 1.797%(w/v) ,KH2PO4 0.497%(w/v) ,therefore,enzyme activity was increased by 18%. The enzyme pecificity was better,corncob being regard as substrate,the optimal enzyme reaction temperature was 65℃ and the temperature was more stable,the optimum pH 6.8 and pH stability was better.
