Preparation of squid ink polysaccharides and its protection against chemotherapeutic injury on testis

Squid ink polysaccharides(SIP),a newly found marine bioactive glycosaminoglycon has been recently proved to have antioxidant ability and chemotherapy-protective activity on model animals.In this paper,a type of SIP containing 3% sulfate group was harvested from squid ink with protease digestion and Sephadex G-100,the molecular weight was about 37ku based on HPLC.Male Kunming mice exposed to chemotherapeutic drug cyclophosphamide(CP) were administered with SIP by gavage,which was used to assess the interventional effects of SIP as a cytoprotector on chemotherapy associated damage in murine testicular tissue.The results showed that SIP alleviated the decrease of mice weight(p 0.01),testis weight(p 0.01) and testis index(p 0.05) caused by CP,and markedly elevated the activity of LDH(p 0.05) and reduced the activity of γ-GT(p 0.01) in testes of CP-treated mice,which indicated significant chemotherapy-protective effects of SIP on murine testis.
