Function of Adrenomedullin in Myocardial Cytoprotection Induced by Exercise Preconditioning

Purpose: To study the function of exercise preconditioning(EP) in protecting rat's myocardial cells from injury caused by isoadrenalin(ISO) and to inquire into the mechanism of function of adrenomedullin(ADM) in myocardial cytoprotection Induced by EP.Methods: Sprague-Dawley rats(n=50) were randomly divided into a control group(C group),an exercise preconditioning group(EP group),an isoadrenalin group(ISO group) and an EP plus ISO group(EP + ISO group).Both of EP group and EP+ISO group performed intermittent treadmill training for three weeks in order to establish an EP model.ISO group and EP+ISO group were injected with ISO into abdominal cavity so as to reproduce the model of myocardial cells injury.The level of cardiac troponinl(cTnl) in serum was measured with immunchemiluminescent method.The change of myocardial cells ultrastructure was observed by transmission electron microscope.The expression of ADM and ADM mRNA in cardiac muscle was showed by immunihistochemistry and hybridization in situ.Results: The cTnl level of EP+ISO group was significantly higher than that of C group,but remarkably lower than that of ISO group.The myocardial cells ultrastructure injury of ISO group was serious,this kind of injury of EP+ISO group was not as serious as that of ISO group.EP group's expression of ADM and ADM mRNA in cardiac muscle was obviously stronger than C group's,while the former's expression of ADM in endothelial cells and smooth muscle cells of coronary vessels was weaker than the latter's.Conclusion: EP makes the expression of ADM in cardiac muscle changed,which protects myocardial cells from ischemia injury induced by ISO.
