Two Drugs Stimulate γ-Globin Gene Expression in K562 Cells

Two drugs,Anisodamine hydrochloride (AAHC) and Harringtonine (HRT), are able to induce an increase of the expression of γ-globin gene in K562 cells. After 2 days of induction by AAHC (10-3mg/mL) and HRT (10-4mg/mL) , benzidine positive cells were increased by 3 fold, with the hemoglobin contents of 2. 16pg and 2. 10pg per cell respectively,in contrast to 0. 41pg per cell of the control. The amount of γ mRNAs were also increased by 50% and 40% above the level found in the control respctively after AAHC and HRT induction. In addition,we have analysed the DNA methylation in K562 cells treated with AAHC and HRT,and the result suggests that the mechanism leading to an incrase of γ gene expression may be associated with an erythroid differentiation.
