Detection of PAGE before SNP Typing

Objective:To explore the role of polyacrylamide gel electrophoresis (PAGE) before conducting single nucleotide polymorphism (SNP) typing.Methods:The PCR products of three DNA fragments (fragment E1 and E2 of DAOA gene,and fragment S4 of RGS4 gene) were detected by PAGE,then compared with the results obtained from denaturing high performance liquid chromatography (DHPLC).Results:The PAGE could clearly distinguish the homozygote and heterozygote of the same SNP site on these three DNA fragments,fragment E1,E2 and S4,as well as two kinds of different homozygotes of the SNP site on fragment S4.Conclusion:PAGE can improve the efficiency of SNP typing,and it is simple and economical.
