Study on Mitochondrial DNA D-loop Sequence Variation in Przewalski's Horse(Equus przewalskii)

Objective:To investigate the genetic diversity and genetic data of Przewalski's horses from Wild Horse Breeding Center in Jimsar county, Xinjiang Province, China. Method: Sequence polymorphisms in 15 individuals of Przewalski's horses mtDNA D-loop region were determined by PCR production direct sequencing. Result: The mitochondrial D-loop hypervariable segment sequences 15464~15866 (402bp) in 15 individuals of Przewalski's Horse were sequenced.12 haplotypes were identfied in the samples, with 37 sites showing polymorphism, it accounts for 9.2% in total sequences, which were 24 transitions, 20 transversion, 8 coexistent sites of transition and transversion and 3 deletions. The contents of A%+ T% 56.1 % was higher than C%+ G% 43.9%. The contents of A, T, G and C were 28.4%,27.7%, 14.9% and 29%, respectively. The haplotypes diversity is 1±0.00116. The nucleotide diversity is 2.90%. The average genetic distance of haplotypes is 0.030. The average nucleotide mutational rate of mtDNA D-loop varied region in 15 individuals of Przewalski's horses was 2.48%. Conclusion: The results showed that the Przewalski's horses maintain high variation in mitochondrial D-loop sequences.
