Optimization Culture of Vibrio harveyi EcGY020401

Objective:It was offered data to produce bacterin of V.harveyi.Method:It was cultured by vibrating or fermenting,CFU/mL of culture was mensurated by plate counting.Result:The results were showed that the NaCl content of 20-25g/L was the most suitable for the growth.The most suitable pH and the glucose content for the growth were 7.5-7.7 and 2-5g/L respectively.The component of TSB medium was optimized,The component of medium improved:tryptone 5 g/L,peptone 15g/L ,soyptone 3g/L,yeast extract 1g/L,glucose 4g/L,K2HPO4 5g/L,NaCl 15g/L,pH 7.5,It was 2.95×1010cfu/mL for cultured strain of EcGY020401,It was needed 10 h to attain the most biologic capacity with 5L ferment vessel,maximum concentration of 3.46×1010cfu/mL.Conclusion:It was cultured strain of EcGY020401 to use TSB medium improved with ferment vessel.it could be reduced cost and increased culturing efficiency.
