Screening and identification of lactic acid bacterium with antimicrobial activity and its biological characteristics

[Objective]The study was to separate and screen lactic acid bacterium with high antimicrobial activity,and to study biological characteristics of antimicrobial substances.[Method] A strain with antimicrobial activity was isolated from fermentation products by plate agar hole diffusion method,and was analyzed and identified by morphological,physiological and biochemical characteristics as well as 16 S r DNA sequence analysis. The antibacterial spectrum was determined. The sensitivity of antimicrobial substances to p H,temperature and five common proteases was investigated.[Result]The strain was identified as Lactobacillus casei,and its fermentation product had inhibitory effect to the bacteria,had no effect on mould and yeasts. The antibacterial zone diameter of the fermentation broth reached 21. 24 ± 0. 45 mm at p H 2. 0,32. 7% higher than that at p H 6. 0. After boiling water bath,the antibacterial zone diameter of the original fermentation broth decreased by 34. 3% and only by 6. 9% at p H 2. 0. After treated with protease,the diameter of the inhibition zone did not change much compared with the control group.[Conclusion]The selected lactic acid bacterium has good antimicrobial activity. The antimicrobial activity and thermal stability of the antimicrobial substances were higher in the acidic condition,and is not sensitive to five proteases.
