The effect of electroporation conditions on the transformation efficiency of Escherichia coli XL1-Blue

The aim of the test is to find out the optimal protocol for electroporation of E.coil XLl-Blue. Changing some important effecting factors, like voltage,concentration of plasmid, the cycle of germ growth et al to find out the optimal protocol. The analysis results showed that the optimal conditions are capacitance 25 #F, electric resistance 200#, voltage 2.5 kV, D600 0.3-0.4, cuvette with an interelectrode distance of 0.2 cm, concentration of plasmid 0.1 #g/ml, cell concentration 2.5#1012, aminobenzylpenieillinare concentration 50 #g/ml. Transformation efficiency is 7.64#108. Electroporation can get high transformation efficiency and repeatability. It lay the foundation for construction of phage display library.
