Effect of Glucose Concentration on Cultivation Process of Transgenic Anabaena sp.PCC IB02 Harboring with Human Tumor Necrosis Factor-α

Objective:To improve the biomass of transgenic Anabaena sp.PCC IB02 harboring human tumor necrosis factor-α(hTNF-α) gene and the expression efficience of hTNF-α by the method of adding the glucose to the BG-11(-N) medium.Methods:Mixotrophic cultivation was carried out in flasks with glucose concentrations in the range from 0 to 300 mmol/L in BG-11(-N) medium.The turbidimetric method and enzyme-linked immunosorbent assay(ELISA) method were used to measure the biomass and the expression efficience of hTNF-α.Results:The data showed that the cell density increased to 3.5 times of that in control sample,and the expression rate of hTNF-α increased to 2%,from 0.5% in no glucose.Conclusion:At different glucose concentrations,the glucose consumption was not obviously.
