Optimization of the lipase imprinting with fat acid in silane matrix

In this study,enzymatic characteristics including esterification activity and alchohol tolerance of the lipase PS effected by fat acid with the carbon chain varied from 6 to 14 was checked.Shown by the results,the molecular similarity with substrate and the hydrophobic property of the imprinting molecules were dramatically affected the activity of the enzymes.With the increasing of the hydrophobic property and the similarity with the substrate,the imprinting effect become distinguished.In imprinting procedure,washing with isooctane keep the highest activity,and the methanol tolerance of imprinted lipase is decrease with the length of carbon chain.The relative activity of imprinted lipases keep 0.65 after used for 5 cycles.The surface scanning electrophic photographs showed that fat acid affected the activity of the enzymes mainly through interferring the structure of the silane matrix.
