The linkage analysis of HLA-DRB_1 and polymorphisms of HLA-DM gene

Objective:To investigate the genetic polymorphisms of HLA-DM genes and to examine whether HLA-DRB1 alleles are in linkage disequilibrium with it. Methods:The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method was used to determine genetic polymorphisms of HLA-DMA/DMB in 106 unrelated healthy Han population from Guizhou province. HLA-DRB1 allele types were examined by using PCR-SSP (polymerase chain reaction-sequence specific primers). Results: The frequencies of DMA0101, DMA0102 and DMA0103 alleles are 0.745, 0.231 and 0.024, respectively. The frequencies of DMB0101, DMB0102, DMB0103 and DMB0104 alleles are 0.547, 0.165, 0.217 and 0.071, respectively. The common genotypes of DMA are DMA0101/0101 and 0101/0102. The common genotypes of DMB are DMB0101/0101, 0101/0102 and 0101/0103. There was linkage disequilibrum between DMA·0102 and DRB1·12. Conclusion: These results showed that the genetic polymorphism of HLA-DM gene in Han individuals from Guizhou Province is distinctive. There was linkage disequilibrum between DMA·0102 and DRB1·12.
