Protective Effects of Ge-132 on Hepatic Ischemia and Reperfusion Injury in Rats

Objective To evaluate the protective effects of Ge-132 on liver injury during normothermic ischemia and reperfusion.Methods The animal model of hepatic ischemia/reperfusion was produced by operation and forty Wistar rats were divided randomly into 4 groups(n=10):A.Normal group,B.Control group,C.Ge-132(25 mg/kg,iv) pretreated group and D.Radix salviae miltiorrhizae(6 g/kg,iv) pretreated group.The hepatic tissue samples were collected to determine the contents of malondiadehyde(MDA),activity of SOD and GSH-PX,activity of Na+、K+-ATPase and Ca 2+-ATPase,and pathologic examinations of the hepatic tissue were made by optical microscope.Results In pretreated groups,hepatic tissue contents of malondialdehyde(MDA) reduced markedly(P0.01),the activity of SOD,GSH-PX and ATPase(Na+、K+-ATPase,Ca 2+-ATPase) in hepatic tissue increased significantly in comparison with the untreated control group.The hepatic injury of pretreated groups lightened evidently.Conclusion Carboxyethyl germanium sesquioxied(Ge-132) attenuate the hepatic injury during normothermic ischemia/reperfusion,which inhibites production of oxygenic free radicals and prevents hepatocellular Ca 2+ overload.
