A Study on Eradicating Virus in Gladiolus by Thermally Treated Meristem-tip Culture

The meristemmed-seedlings were tested for viruses by biological tests, counterimmunoelectrophoresis,indirect enzymelinked immunesorbent assay and immune electronmiroscopic methods. It was shown that the efficiency of eradicating virus by heat treatmentwith meristem-tip culture was higher than that by meristem-tip culture alone, especially in thetreatment of 42 ℃ for 5 days. In addition, the culture medin were improved in order to fit todedifferentiation of explantlets of Gladiolus. The cycle of the tissue culture was greatly shortened.As a redult of this, one apical stem explant could propagate more than 30000 times in oneyear.The procedure of producing virus free come of Gladiolus was proposed for commercialusage.
