Effect of Curcumin on Behavior of Autism Rats and the Expression of Brain Derived Neurotrophic Factor

Objective To study the effect of curcumin on behavior of autism rats and expression of brain derived neurotrophic factor(BDNF).Methods Animal model of autism was established according to Schneider method,the female that received a single intraperitoneal injection of 600 mg·kg-1 sodium valproate on the 12.5 day after conception and then the auxanology,behavioral and morphological test were performed to identify autism mouse.Randomly selected animal model of autism(n=20) and offspring of the NS group(n=10)at age of 35 days,the saline group rats were designated as normal control(Con)group,the animal model of autism was randomly divided into vehicle-control group(VC group,n=10)and curcumin group(VPA-C group,n=10),the VPA-C group was injected curcumin into abdominal for 2 weeks(concentration of curcumin was 10 g·L-1 which was dissolved in PBS containing 1 mL·L-1 dimethyl sulfoxide).The solvent control group and normal control group were given PBS with volume of 1 mL·L-1 DMSO for 2 weeks.This investigation was observed the effects of curcumin on rat behavior and used the immunohistochemical method to observe the expression of BDNF in the different groups.The results were statistically assessed by SPSS 18.0 software.Results The effects of curcumin on autism rats were starting to increase the number of social behavior(P0.05),to decrease latency to social behavior(P0.001),and to reduce repeat behavior of autism mouse(P0.01);Curcumin increased BDNF integral optical density values of autism rats in temporal cortex(P0.05).Conclusions Curcumin have a therapeutic effect on rats with autism,it can be improved abnormal behavior of autism rats to some extent,the mechanism may be to play a role by increasing the expression of BDNF.
