Determination of artemisinin and its related sequiterpenoids in leaves of Artemisia anuua at full flowering by HPLC-UV-ELSD

Objective To establish an HPLC-UV-ELSD method for determination of artemisinin and its related biosynthetic precursors in leaves of Artemisia annua.Methods Venusil XBP-C18 column(200 mm×4.6 mm,5 μm) was applied.Artermisinic acid analyses were performed on a HPLC chromatograph with UV detector.H2O/formic acid/CH3CN(V∶V∶V=40.00∶0.08∶60.00) was used as mobile phase,the detection wavelength was set at 215 nm.Other three related analytes were determined on UV detector and ELDS detector.The carriergas pressure was 50.1 psi,and parameters of ELSD were set as S.C.35 ℃ and D.T.60 ℃.Gradient elution(CH3CN/H2O) was carried out with detection wavelength of 203 nm.Results Standard calibration curves,constructed forartemisinin and its biosynthetic precursors,were linear between 0.10 and 1.12 g·L-1(r≥0.999 0).The average recoveries were 98.5%,96.5%,97.5%,95.4%,with RSD of 2.3%,1.8%,3.6%,2.5%,respectively.Conclusions The method established in the study was convenient,reliable and accurate,and it can be used to monitorthe contents of artemisinin along with its biosynthetic precursors in leaf samples of A.annua during the vegetation period.
