The anti-hepatitis B virus activity of NAC in vitro

Objective To explore the anti-hepat iti s B virus (HBV) activity of NAC in vitro. Methods HBV-produ c ing cell lines HepG2-2.2.15 was treated with various concentration NAC (0,3,10, 3 0 mmol/L). Hepatitis B Surface antigen (HBsAg) and hepatitis B e antigen (HBeAg) was measu red by the ELISA. Semiquantity-PCR was used to analyze the quantity of the HBV D NA in the cell and the culture supernatant.The mRNA of the hepatitis B Surface a ntigen (HBsAg) in the cell was analyzed by RT-PCR. Results N A C is able to inhibit HBsAg and HBeAg at a dose-dependent and time-dependent ma nn er, however NAC inhibits HBsAg more effectively. The treatment of HBV-producing cell lines with NAC results in a dose-dependent reduction of the HBV DNA in the culture supernatant,but the quantity of the HBV DNA in the cell arise with the i ncrease of the NAC concentration. The mRNA of the hepatitis B Surface antigen (H BsAg) in the cell was not affected. Conclusion NAC exerts its anti-HBV activity at a posttranscriptional level.
