Expression of macrophage migration inhibitory factor (MIF) and hepatocyte growth factor (HGF) in nasopharyngeal carcinoma andits relation to clinical prognosis

ObjectiveTo investigate the expression of macrophage migration inhibitory factor (MIF) and hepatocyte growth factor (HGF) in nasopharyngeal carcinoma (NPC), and its correlation with the clinical prognosis of NPC. MethodsForty-five tissue specimens were collected from biopsies of patients with NPC during the period of 19992003. Immunohistochemical staining was used for detecting the expression of MIF, HGF-α in NPC tissues. The number of intratumoral microvessel density (IMD) was evaluated by counting the CD34-positive neovessels in the most active areas of tumor neovascularization or "hotspots". The associations of these indexes with clinicalopathological features and follow-up data were analyzed by Kaplan-Meier method. ResultsIn 45 cases, the positive expression rate of MIF in tumor cells was (77.8% (35/45).) However, only 1 case (2.2%, 1/45) showed HGF positive signal in neoplastic cells. HGF was abundantly detected in the interstitial tissues surrounding NPC, in which lymphocyte was HGF-positive predominantly. The expression level of MIF and IMD count showed a correlation with lymph node metastasis. NPC tissues with lymph node metastasis had higher expression of MIF and higher IMD count than those without lymph node metastasis (P=0.029 and 0.026,respectively). Meanwhile, the number of IMD counted in MIF positive group was significantly higher than that in MIF negative group (35.13.3 vs 20.910.2/HPF,P=0.003). The number of IMD counted in higher HGF expression group [CM(53*3]significantly increased, compared with lower expression one ((36.5)(15.6) vs (26.2)(12.4)/HPF,P=(0.011)). Kaplan-Meiersurvival curves described that the survival was significantly lower in NPC patients with both high MIF and high HGF expression (P=0.033). For patients with high IMD, the survival also decreased,compared with lower IMD patients (P=0.048). ConclusionsMIF and HGF are closely associated with tumor neovascularization and the higher expression of MIF and HGF synchronously in NPC might be associated with worse prognosis of patients.
