A NOVEL RAT MODEL OF TYPE 2 DIABETES MELLITUS

The study was aimed at developing a rat model that closely simulates the metabolic abnormalities of human type 2 diabetes mellitus. METHODOLOGY Female Wistar rat was fed with a diet enriched with sucrose (10%,w/w),triglyceride(10%,w/w)and cholesterol(5%,w/w)to induce insulin resistance,and after one month on the diet streptozotion(STZ,25 mg/kg)was injected intraperitoneally to induce hyperglycemia. RESULTS Insulin levels increased in response to the diet(33 5±1 97 mU/L vs 18 3±2 04 mU/L in controls, P 0 01)and insulin resistance was confirmed by insulin suppression test.After STZ injection hyperglycemia(15 5±7 80 mmol/L vs 5 70±0 90 mmol/L in controls, P 0 05)was observed,and plasma insulin decreased to levels of chow-fed rats,with hypertension and hypertriglyceridemia and hypercholesterolemia. CONCLUSION This novel rat model simulates human type 2 diabetes in moderate hyperglycemia,hypertension,dyslipidemia and insulin resistance.It may provide a particular advantage for investigating diabetic vascular complications and pharmacology.
