Study on Extraction Process of Polysaccharide from Dendrobium fimbriatum Hook.var.oculatum Hook by Ultrasonic Assisted Hot Reflux

Objective To optimize the extraction process of polysaccharide from D.fimbriatum Hook.var.oculatum Hook by ultrasonic hot reflux assisted.Methods On the basis of single factor experiments,the extraction process was optimized by orthogonal experimental design on effects of extraction temperature,ultrasonic-irradiation time and extraction times.The yield of polysaccharide was used to evaluate the factor levels.Results The optimum conditions were determined as follows: ultrasonic-irradiation 16 min at 95℃ and extraction four times.The extraction rate of polysaccharide reached(18.64±0.40)% by using the method,which was much better than hot reflux extraction method.Conclusion That the method is simple with high accuracy,good stability and reproducibility,and can be used to extract polysaccharide from D.fimbriatum Hook.var.oculatum Hook.
