Determination of the Contents of Total Flavonoids in Elaeagnus angustifolia Blossoms and its Pharmacological Actions

Objective To determine the contents of total flavonoids and rutin in Elaeagnus angustifolia blossoms(EAB)according to Technical Standard For Chinese Pharmacopoeia(2005 version),and to study the protective effects of the alcohol extract of Elaeagnus angustifolia blossoms(AE-EAB) on acute hepatic injury induced by carbon tetrachloride(CCl4) in mice.Methods Spectrophotometric method and high performance liquid chromatography were used to determine the contents of total flavonoids and rutin in EAB,according to Technical Standard For Chinese Pharmacopoeia(2005 version).The levels of ALT,AST in serum and MDA,SOD and GSH in liver were measured and the hepatic histological changes were observed by optical microscope in order to study the protective effects of different doses of alcohol extract of AE-EAB on acute hepatic injury induced by 0.1% CCl4 in mice.Results The content of total flavonoids of EAB is 27mg/g(27mg total flavonoids in 1g EAB),and that of rutin was 0.029 mg/g(0.029 mg rutin in 1g EAB).Compared with model group,the levels of ALT,AST in serum were obviously decreased(P0.05),and the level of MDA in liver was also lowered(P0.05),but the activities of GSH and SOD in liver were markedly improved(P0.05,P0.01) after pretreatment with AE-EAB.AE-EAB ameliorated the hepatic pathological changes too.Conclusion The results show that EAB contains flavonoids,and AE-EAB has protective effects on acute hepatic injury induced by CCl4 in mice.
