TTF1-induced Apoptosis of HepG-2 Cells through A Mitochondrial Pathway

Objective To study the induction on apoptosis of 5,2',4'-trihydroxy-6,7,5'-trimethoxyflavone(TTF1) in human hepatocellular carcinoma HepG-2 cells and its mechanisms. Methods HepG-2 cells were incubated in vitro in the presence of TTF1,cell cycle and apoptosis were observed by using flow cytometry.The protein expressions of bcl-2,bax,cyt-c,caspase-3 and caspase-9 were assayed by using western blot. Results TTF1 both inhibited cell growth and induced apoptosis.Western blot analysis showed that TTF1 treatment led to decrease of transcription and protein expression of bcl-2 and increase of bax,Cyt-c caspase-3 and caspase-9. Conclusion TTF1 may induce apoptosis of HepG-2 cells through a mitochondrial pathway.
