Determination of Chlorogenic Acid in Tea by High Performance Liquid Chromatography

A sensitive and precise method has been developed for the detection and quantitation of chlorogenic acid in tea. The chlorogenic acid was seperated from methanol extract of tea by RP _ HPLC with pecosphere C18 column and detected with UV detector at 326 nm. The mobile phase consisted of solvent A(methanol) and solvent B(water including 1/61(φ) formic acid). The column was eluted with following gradient: 0～17 min 18% A; 18～27 min 100% A. The calibration curve is linear in the range of 0.02～4.02 μg for chlorogenic acid. The spiked recovery rates are 86.8% and 102.0% for Gongju tea in Huang Shan and Honeysuckle tea in Anhui, respectively. The method has been applied to practical samples over 10 kinds of tea.
