Isolation,Purification and Preliminary Structure Analysis of Polysaccharide DCPP3c-1 in Suspension-cultured Protocormns of Dendrobium Candidum

Crude polysaccharide DCPP(polysaccharide of protocorms of D.candidum)was isolated and purified by ion-exchange chromatography on DEAE-52 column and gel filtration chromatography on Sephadex G-200 column.The purity of the resulting gray polysaccharide component DCPP3c-1 was verified by specific rotatory power test,Sephadex G-200 chromatography,and UV spectra scanning.Its composition and structure were analyzed by TLC,HPLC,IR spectra and periodate oxidation.The results indicated that DCPP3c-1 was a single homogeneous component,and its average molecular mass was estimated to be 72.4 ku.Mannose,rhamnose,galaduronic acid,glucose,galactose and arabonose were found in DCPP3c-1 with a molar ratio of 1.120 4 ∶1 ∶1.046 ∶23.354 ∶3.828 ∶1.046,respectively,and it also consisted of α-pyranoside with(14% of 1→6,40.7% of 1→2 or 1→4,45.3% of 1→3)glycosidic linkages.Meanwhile,the IR spectra showed that it had characteristic absorptions of polysaccharide and α-glycosidic bond.DCPP3c-1 was a novel acidic heteropolysaccharide,and this was the first report of its isolation and purification from suspension-cultured protocormns of D.candidum.
