Construction of SSR-based Molecular Finger Printing and Analysis of Genetic Diversity for Lonicera japonica

SSR markers were used to construct the fingerprinting and genetic diversity of Lonicera japonica.25 primer pairs were synthesized to test the amplification and polymorphism in 6 Lonicera japonica cultivars.The results showed that 3 primer pairs presented stronger polymorphism.The Excel-based digital fingerprinting was established.UPGMA cluster analysis was used to estimate the genetic distances and to construct a dendrogram by SLT-NTSYS.The results showed that the coefficient of genetic similarity of Lonicera japonica and Lonicera confusa was only 0.28.87 commercial samples were clustered using the SSR fingerprinting,and divided into 3producing areas as expected. The Lonicera japonica SSR fingerprinting was reliable and convenient,and could be used in germplasm identification.
