An Efficient Expression System of Recombinant Human Interferon-α 2b in E.coli

Objective: To construct the prokaryotic expression system of recombinant human interferon-α2b(rhIFN-α2b) to receive its efficient expression in E.coli. Methods: The artificially semi-product was suitable to expression of coding sequence for rhIFN-α2b in E.coli according to the frequency table of gene codon in E.coli and unchanged the conformation of amino acid.The expression system was amplified by PCR then coloned into expression plasmid PDH and positive bacteria clutched which were selected performed temperature induced expression and the expression products were applied to SDS-PAGE,Western Blot identification and WISH-VSV system to identify anti-virus vitalism. Results: The anticipated 512 bp DNA strand was observed by using jelly gel to identify PCR products,recombinant plasmid pDH/IFN-α2b which contained the anticipated and correct reading frame rhIFN-α2b codon sequence was detected by using sequence analysis ,the protein strand which had the same molecular quality to rhIFN-α2b observed by SDS-PAGE and this strand identified by Western Blot and its specific activitity was able to obtain (1.8~2)×108 U/mg. Conclusion: The efficient expression system of rhIFN-α2b in E.coli is constructed successfully and the biological vatalism can be expressed greatly.
