To optimize the supercritical CO_2 extraction process of volatile oil in Flos lonicerae and to study the anti-inflammatory activity of the extracted product, the influences on temperature,pressure,CO_2 flowing and extracting time to the volatile oil by supercritical CO_2 extraction were observed. The optimum extraction was investigated by the orthogonal design.Anti-inflammatory activity was studied by mouse ear swelling model. The optimum extracting conditions were: the temperature 35 ℃,the pressure 12 MPa,the CO_2 flowing 4.0 kg/h and time 1.5 h.The extraction rate was 1.08 % and as 43.2 times high as that by the steam distillation method. The supercritical CO_2 extracting volatile oil was of potent anti-inflammatory effect on the ear swelling model in mice. This optimized process is simple, stable, efficient and the anti-inflammatory effect of the extracted oil is potent.
