Analysis of Anthraquinones in Different Samples of Aloe with HPLC-DAD and LC-MS/MS

A HPLC-DAD and LC-MS/MS method was established not only for the assay of anthraquinones but also for the determination of aloin and aloe-emodin in different samples of aloe.The analysis was performed on Nucleodur-silica C18(250 mm4.6 mm,5 μm)column with a methanol-water(containing 0.4% acetic acid) gradient at ambient temperament.The flow rate was 1.0 mL/min,the wavelength scanned from 190-370 nm and the detection was at 254 nm and 356 nm.The ionic source of LC-MS was ESI.Samples were analyzed in two modes:MS and MS/MS full can modes.Aloin A,B and aloe-emodin were the main anthraquinones in different samples of aloe while rhein,emodin,chrysophanol and physcion were not found;the aloe powder contained the highest amount of aloin.The method could be used to control the quality of aloe product.
