Distribution of ~(99)Tc~m-rh-Annexin Ⅴ in Tumor and Expression Relationship of bcl-2, bax After a Single Dose of Chemotherapy

The expression of bcl-2 and bax after the single dose of chemotherapy with 99Tcm-rh-AnnexinV as the tracer of tumor apoptosis imaging is studied. Tumor cell apoptosis is examined by TUNEL methods, and the expression of bcl-2 and bax in tumor are determined by immunohistochemical methods. Single dose of chemotherapy significantly increased the tumor uptake of 99Tcm-rh-annexinⅤ and the positive number of TUNEL, as well as the expression of bax (P0.01). The expression of bcl-2/bax is significantly higher in contrast group than in therapy group (P0.01). The radio-uptake in tumor correlates well with the positive number of TUNEL in contrast group and therapy group(r=0.801, r=0.769). In therapy group, the expression of bax correlates well with the uptake and the positive number of TUNEL(r=0. 849, r=0.652), but the bcl-2/bax correlates negatively with radio-uptake and the positive number of TUNEL(r=-0. 820, r=-0. 694). The distribution of 99Tcm -rh-annexin Ⅴ in tumor reflectes not only the degree of apoptosis of tumor cells, but also the change of bax expression after the single dose of chemotherapy.
