Increased gene expressions of Apelin and its receptor in 2K1C renal hypertension rats

Objective:To examine the expression changes of apelin and its receptor (APJ) genes in left ventricle tissues of two-kidney, one-clip(2K1C) rats. Methods: Twenty male Sprague-Dawley rats were randomly divided into normal control group and model group. The mean of carotid arterial pressure (mCAP), mean of left ventricular end diastolic pressure (LVEDP), the maximum rate of change in left ventricular pressure(LV±dp/dtmax), and the weight ratio of left ventricular mass (left ventricle plus septum)to bodyweight (LVM/BW) were measured to estimate the model of 2K1C renal hypertension. The expression changes of apelin and APJ genes of left ventricle (LV) were detected with reverse transcription- polymerase chain reaction (RT-PCR). Results: ①The mCAP, LVEDP and LVM/BW of 2K1C group were higher (36.58%, 333.8% and 20.24%,respectively) than those of normal control group (P0.05, P0.01, P0.01), but LV+dp/dtmax was lower 85.35%(P0.05). However LV-dp/dtmax was of no significant difference between two groups. ②The expressions of apelin and APJ mRNA in left ventricle tissues of 2K1C rats were higher (77.66% and 119.00%,respectively) (all P0.05) than those of normal control group. Conclusion: The local increased expressions of apelin and APJ genes in left ventricle in renal hypertension rats may play an important role in the development of left ventricular hypertrophy.
